Extraction Method of Rosemary Extract

Publish Time: 2023-02-24     Origin: Site

The main components of Rosemary Extract are rosemary phenol, syringeal, and syringic acid. Several main components in Rosemary Extract have strong antioxidant activity. It has significant effects in preventing the oxidation of fats and oils and maintaining the meat’s flavor. Let's take a look at the extraction method of Rosemary Extract.

The following is a list of contents:

  • Organic solvent extraction method

  • Ultrasound-assisted extraction

  • Supercritical carbon dioxide extraction

Organic solvent extraction method

The organic solvent extraction method is generally used for the extraction of active ingredients in herbal medicines and is the most commonly used extraction method. The main chemical components in rosemary, mono steroids, and sesquiterpenoids, and their derivatives are fat-soluble substances, insoluble in water, so this characteristic can be used to extract with methanol, ethanol, ethyl acetate, and other organic substances as solvents, because ethanol is cheap, non-toxic, easy to recover, and it is reported that the extraction rate of ethanol is higher than other organic solvents, so ethanol is often chosen as the extraction solvent. This method is easy to operate, requires simple equipment, and has high product yield, but at the same time its cost is higher, the impurity content is higher, the extraction time is longer, and it is very easy to lead to the solvent residue, which must increase the distillation device to remove the organic solvent, and in the distillation process may lead to the loss of volatile components, so it is not suitable for industrial production of Rosemary Extract.

Rosemary leaves were crushed and extracted with ethanol, then the extract was mixed with glycerol esters of C6~C12 fatty acids, ethanol was removed by rotary evaporation, hexane was added for dilution, insoluble matter was filtered out, and hexane was removed by rotary evaporation to obtain Rosemary product, which contained about 5% of antioxidant components. Rosemary Extract was extracted by reflux method using organic solvents, and the process conditions of Rosemary Extraction were optimized by single-factor and orthogonal experiments, and the content of the active ingredients of Rosemary Extract was determined.

Ultrasonic-assisted extraction method

Ultrasound can produce high speed and powerful physical action to destroy the cell wall of plant cells, and also has a certain agitation effect, so that more active ingredients in the cells flow out and dissolve into the solvent, which effectively improves the extraction efficiency, the extraction time is shorter than other methods, saving the solvent, and at the same time, this method operates without heating, avoiding the destruction of active ingredients by high temperature. Due to these advantages, in recent years, people are increasingly using the ultrasonic-assisted method for the extraction of active ingredients of herbal medicines, making it a relatively common Rosemary Extract extraction method.

Supercritical carbon dioxide extraction method

The supercritical carbon dioxide extraction method is when the carbon dioxide gas is in the critical condition of liquefaction, the extracted substance is contacted in the reaction vessel, and the solubility of the extracted substance is changed by the great pressure and extremely low temperature in the vessel, so that the active ingredients are extracted separately according to the different polarity size and boiling point, thus achieving the purpose of separation and purification. By using the supercritical CO2 extraction method, the extraction efficiency can be greatly improved, there is no organic solvent in the whole process, so there is no solvent residue in the product, which reduces toxicity, and the whole process is carried out at a low temperature, so there is no high temperature to destroy the extract. Not only can the functionality of the extracted substance be preserved to the maximum, but also the separation can be targeted. When extracting non-volatile substances, a certain amount of entrainment agent should be added for better extraction of Rosemary Extract.

It has been shown that using ethanol as an entraining agent, the raw material of Rosemary is placed in a reactor, the pressure and temperature are set, and the antioxidant of Rosemary is obtained through carbon dioxide using a supercritical apparatus, in which the main component is syringic acid. Rosemary Extract was obtained by supercritical extraction with a high concentration of ethanol as an entraining agent, and the content of fat-soluble and water-soluble active ingredients was determined by using rosemary as raw material under certain pressure and temperature conditions, and the yield was 11.93% in general. The same supercritical extraction method was used to extract syringic acid from rosemary with the help of carbon dioxide, and the purity of syringic acid obtained was 37%.
